Antitumour Functions of γδ T Cells

Although γδ T cells constitute a minor subset (1-5%) of total circulating T cells, they possess a combination of innate and adaptive immune cell qualities. In immunotherapy, γδ T cells exert direct/indirect antitumor cytotoxicity, produce inflammatory cytokines, directly lyse infected or malignant cells, and establish a memory response to attack pathogens upon re-exposure.

  • Direct tumor cell targeting by γδ T cells
  • γδ T cells play a direct antitumor role mainly from four different aspects: (1) directly lyse cancer cells through the perforin-granzyme pathway; (2) eliminate cancer cells through the ligands TRAIL and FasL that can enhance the susceptibility of the cancer cells to γδ T cell lysis; (3) kill cancer cells via antibody-dependent cell-mediated cytotoxicity (ADCC); (4) γδ T cells are important early sources of IFN-γ and TNF-α to inhibit cancer growth.

  • Indirect effects οf γδ T cells on antitumor immunity
  • γδ T cells exert their indirect antitumor effect by orchestrating downstream immune responses. They can interact with B cells, DCs, αβ T cells, and NK cells to participate in tumor immune regulation. For example, γδ T cells have been shown to affect B cell antibody secretion in non-immunized mice. In addition, they can also act as antigen presenting cells (APCs) for αβ T cell priming. DC maturation and robust NK cell-mediated antitumor cytotoxicity are also associated with γδ T cells.

γδ T Cells and Prostate Cancer

Prostate cancer is a common type of cancer in males that affects thousands of men each year. It has been the second-leading cause of cancer death for men in the United States. Protective effects by γδ T cells have been validated in models of B cell lymphoma, colorectal cancer and prostate cancer. Liu et al. (2008) used a transgenic mouse model of prostate cancer (TRAMP) to firstly reveal that γδ T cells can provide protective immunosurveillance against spontaneously arising mouse prostate cancer. TRAMP is a model of spontaneously arising mouse prostate cancer that mimics human disease. Therefore, these findings provide scientific evidence that γδ T cells provide protective innate immunosurveillance against prostate cancer, and they provide a rationale for designing adoptive transfer of ex vivo expanded autologous γδ T cells to treat prostate cancer.
