Technologies for γδ T Cell Isolation

Magnetic bead cell sorting (MACS) is a common, efficient, and fast method to isolate untouched γδ T cells from human peripheral blood mononuclear cells (PBMCs). The basic principle is to deplete non-γ/δ T cells using magnetic particles labeled by antibodies recognizing specific cell surface markers. Non-γ/δ T cells such as α/β T cells, NK cells, B cells, granulocytes, monocytes, dendritic cells, stem cells, and erythroid cells are indirectly labeled with a cocktail of biotin-conjugated monoclonal antibodies. MicroBeads are used to capture these labeled non-γ/δ T cells. Subsequently, the labeled non-γ/δ T cells are retained on a MACS column in the magnetic field of a MACS separator, while the unlabeled γ/δ T cells pass through the column. These untouched γ/δ T cells are very suitable for subsequently phenotypical and functional analysis, such as studying cytokine expression and receptor signaling, antigen recognition, cytotoxicity.
