It has been documented that the proportions of γδ T cells were increased in the cerebrospinal fluid (CSF) and peripheral blood from PD patients, moreover, patients with PD usually had higher numbers of γδ T cells. These data suggested that γδ T cells may play in PD pathogenesis and also provided new insights into PD therapies.
AD is an incurable neurodegenerative disorder in which neuroinflammation has a critical function. The roles of γδ T cells in AD are gradually understood by deep sequencing of the CDR3 region of T-cell receptor γ genes (TRGs) in patients with AD and control patients. Scientists found that AD was associated with a unique TRG repertoire, and the chemical properties of CDR3 regions may be altered in AD. These findings highlighted the potential of AD-γδ TCR repertoires as new biomarkers for prediction and diagnosis of AD.
γδ T cells can be activated to induce various inflammatory cytokines, such as IFN-γ, IL-4, IL-17, IL-21, and IL-22. In encephalomyelitis cases, γδ T cells function as the major initial producers of IL-17, suggesting their crucial role in the pathogenesis of encephalomyelitis. Additionally, γδ T cells express high levels of IL-23R and IL-1R, which further enhance their effector functions in the pathogenesis of encephalomyelitis.
RE is a rare neuroinflammatory disease characterized by intractable seizures and progressive atrophy on one side of the cerebrum. It was often accompanied by clusters of T cells in the affected cortical hemisphere, indicating an active cellular immune response. Activated γδ T cells are associated with neuroinflammation in RE. In RE cases, the inflammatory factor IFN-γ is increased, which is the product of γδ T cells, suggesting γδ T cells can be considered as a marker of RE as well as the indicators of the choice of therapy for future practice.
