Introduction to γδ TCR Repertoires

γδ T cells are a rare type of T cells that are of great importance in cytokine production, tissue regulation, and immune defense against tumors. γδ T cells express specific γδ TCR that consists of a γ-chain and a δ-chain. The genes encoding these two chains of γδ TCR are rearranged in the thymus. During this process, the variable (V) domain, the diversity (D) domain, and the joining (J) domains of somatic DNA recombine and create the diversity of γδ TCR chains. As a result, all γδ T cells are merged to form large γδ TCR repertoires. Moreover, pilot studies have indicated that the composition of specific γδ TCR repertoires is associated with the disease development in the human body. However, only a few TCR ligands, proteins, or factors have been confirmed that can be used for promoting the γδ TCR repertoire and activation.

In this condition, many attempts have been made to generate novel technologies for analyzing human γδ TCR repertoires, identifying the γδ TCR ligands, as well as illustrating the mechanism of TCR diversity in disease models. For example, high-throughput TCR repertoire sequencing and next-generation sequencing (NGS) technology have become powerful tools in much academic research.

γδ TCR Repertoires Analysis Services

Currently, γδ T cells have been considered as unique innate-like T cells with TCRs that can easily recognition of highly conserved antigens. Meanwhile, evidence has suggested that analyzing the γδ TCR repertoire is important to obtain meaningful data for the immune system features, the causes, and the progression of the disease. At present, the main obstacle to the problem of the γδ TCR repertoire services is that traditional methods are not sufficient to characterize the TCR repertoire in-depth.

To solve this problem, Creative Biolabs has established a panel of assays for the analysis of the γδ TCR repertoire. Currently, we can provide a seamless service chain, ranging from gene expression profiling, γδ TCR repertoire analysis, γδ T cell subsets identification, to development and maintenance of γδ T cell in various disease models. For instance, monoclonal antibodies have been used for detecting specific V gene subgroups of γδ TCR repertoire. A wide variety of polymerase chain reaction (PCR) strategies have been generated for offering a fast and accurate overview of the repertoire.

Besides, next-generation sequencing (NGS) based technologies are most widely employed recently for clonotype identification and γδ TCR repertoire analysis. In our labs, the high-quality RNA libraries from different samples, such as human peripheral blood mononuclear cells (hPBMCs) and purified T cells, will be established to capture the V(D)J variable regions of TCR transcripts and testing the alpha- and beta-chain diversity of γδ TCR.
