CAR γδ T Cell Viability Test

Tetrazolium-based cell viability assays include MTT, MTS, XTT, WST, and CCK-8. These assays are based on the reduced tetrazolium salts in living cells. Tetrazolium salts can be reduced by living cells to form colored formazans. The color change can be measured by absorbance microplate readers.
Resazurin (blue) is a popular dye. It can be reduced by viable cells to form colored resorufin (pink). The change of color is measured in absorbance mode by acquiring absorbance at 570 nm.
ATP as a cell energy source is proportional to the number of living cells. ATP-based cell viability assays are based on the ATP-dependent luciferase reaction.

We support the detection of multiplex cytokine production such as interferon-gamma (IFNγ), interleukin-2 (IL-2), and other cytokines based on enzyme-linked immunosorbent assay (ELISA), Luminex multiplex assays, and bead-based flow cytometry.
The cytotoxicity test is designed to evaluate the killing or toxic effect of CAR-γδ T cells on target tumor cells. We use the co-culture of target tumor cells and CAR-γδ T cells to monitor target cell lysis, effector cell frequency, and activity based on flow cytometric technology. We also use radioactive chromium-release to assess the cytotoxicity of CAR-γδ T cells on target tumor cells.
