Introduction to CAR Engineered γδ T cell

Rischer et al. demonstrated that peripheral blood-derived Vγ9Vδ2 T cells transduced with retroviral vectors encoding either GD2 or CD19-specific CARs had high CAR expression and could be readily expanded. In vitro tests suggested that CAR-expressing γδ T cells might serve as potent and specific antitumor effector cells. Based on these studies, researches and applications of CAR γδ T cells have been in rapid development. Typically, first-generation CARs had a single CD3-z intracellular signaling domain; they were shown to have poor persistence in vivo. Second-generation and third-generation CARs were created by the addition of one or two costimulatory domains to the CD3-z motif to provide tailored signals to enhance activation and survival. Additionally, because CAR γδ T cells can act as professional antigen-presenting cells, combination therapies with other modalities of immunotherapy such as checkpoint inhibitors, oncolytic viruses, vaccines, or cytokines could synergistically amplify recruitment and function of tumor-infiltrating lymphoid and non-lymphoid cells.

While γδ T cells clearly show potent antitumor activity, some reports describe the regulatory function of these cells in the tumor microenvironment.
