γδ T cells are innate immune cells expressing a T cell receptor (TCR). It can express Vδ1 chains which are prominent in the intraepithelial layer of mucosal surfaces, where they are involved in maintaining epithelial tissue integrity when facing damage, infection, or transformation, responding to stress antigens on epithelial cells, and producing interleukin-10 (IL-10). Phenotypic Characterization of γδ T-Cell Subsets

Cytotoxicity of human Vγ9Vδ2 T cells is dependent on the NKG2D activation and is mediated by Fas/Fas ligand (FasL) and perforin-granzyme B pathways. Compared the levels of surface NKG2D and FasL expressions with the intracellular expression of cytotoxic granules for the CD56+ and CD56-Vγ9Vδ2 T-cell subsets, we found that CD56+ and CD56- γδ T-cell subsets expressed with similar levels of NKG2D, perforin, granzyme B, and FasL, as well as CD69, which is a recent activation marker. CD56+ Vγ9Vδ2 T cells expressed a significantly higher level of CD16 than CD56- counterparts.

    The basis of T cell immunophenotyping is that T cells have specific cell surface receptors, intracellular cytokine expression profiles and / or other intracellular proteins with different phosphorylation. This allows these cells to be selectively labeled with specific markers. According to the expression of CD3, the immunophenotype of T cells can be analyzed by immunohistochemistry (IHC) method, and then further subdivided according to other specific markers.
