γδ T cells are involved in immune surveillance against hematological neoplasms and solid tumors and the pathogenesis of autoimmune diseases. Vδ2 T cell responds mainly to intracellular bacteria and solid tumors, while Vδ1 T cell, resident mainly in tissues, is used mostly in the defense against viruses and malignancies. γδ T cells are being explored in cell-based immunotherapy. Enriched γδ T cells can be prepared by magnet separation and the purity of γδ T cells is up to 85% to 90%, as assessed by flow cytometry analysis, and their cell viability is up to 90% as determined by trypan blue exclusion.
Function of Zebrafish γδ T Cells in Vitro
An in vitro antigen-specific CD4+ T cell activation assay was performed to investigate whether zebrafish γδ T cells can act as antigen-presenting cells (APCs) to initiate adaptive immunity. The distribution of major histocompatibility complex (MHC-II), CD80/86, and CD83, three specific APC markers involved in Ag presenting and costimulatory signaling to CD4+T cells, were determined in the activated γδ T cells magnetically sorted from the antigen (KLH)-stimulated fish through reverse transcription PCR (RT-PCR) and double-immunofluorescence staining. This result provides initial evidence that zebrafish γδ T cells exhibit the functional characteristics of APCs.
