Introduction to Lactate Dehydrogenase (LDH) Assay

LDH, a ubiquitously-expressed intracellular enzyme, catalyzes the reversible oxidation of lactate to pyruvate. LDH can release into the supernatant when cell damage or lysis, because this enzyme mostly exists in the cytosol. Hence, LDH is usually used to measure cytotoxicity and cell number in the in vitro cell culture systems. Nowadays, detecting levels of LDH enzyme has become a routine and fundamental method of study the cytotoxicity effects of experimental drug formulations and cell transfection. In Creative Biolabs, our LDH cytotoxicity test enables accurate LDH levels determination in cultured cell supernatants from a wide range of cell lines by using a simple, proven colorimetric (UV) enzymatic assay to specifically detect LDH.

Several previous studies have detected γδ T Cell cytotoxicity through the LDH Cytotoxicity assay. For example, in an analysis of tumor-infiltrating γδ T cells in rectal cancer, the LDH assay was done. In order to detect specific cytotoxicity, HR8348 and Vδ2 T cells were co-cultured at the ratios of 10:1, 20:1 and 30:1. Results showed that the inhibitory effects of Vδ2 T cells from para-carcinoma tissues and from rectal cancer tissues were not significantly different.
